Abstract of "Study of the Lopolytic Activities of Fungi Isolated from Different Oil Mill Areas"
A screening programme concerning the lipolytic fungi of different oil mill areas located at various localities around Kathmandu, Lalitpur, Bhaktapur and Kirtipur were undertaken. Of thirty different fungal isolates, seven showed lipolytic activity when grown in media incorporated with lipid substrate. The optimum culture conditions for maximum lipase production were determined by growing the fungi on solid agar media. All the lipolytic fungi produced higher lipolytic activity in presence of Tween 80 as sole carbon source and yeast extract as nitrogen source. On the other hand, they showed varied response towards temperature and pH. The lipase activity was then studied in liquid submerged culture at two different temperatures of 40 and 30 degree Celsius at pH 6.2. At 40 degree C, the 4 days old cultures of Aspergillus niger and Aspergillus spp showed the best lipase activity of 45.25 µmol of free fatty acids per minute per gram dry cell mass and 63.23 µmol of free fatty acid per minute per gram dry cell mass respectively. Similarly, best lipase activity of 112.15 µmol of free fatty acid per minute per gram dry cell mass and 52.73 µmol FFA per minute per gram dry cell mass were achieved with the fungi Aspergillus fumigatus and Aspergillus flavus respectively under similar culture conditions at 30 degree C. Alternaria alternata and Cladosporium herbarum showed the least lipase activity of 9.69 µmol of FFA per minute per gram dry cell mass and 2.11 µmol of FFA per minute per gram dry cell mass respectively at 30 degree C.
